Two oyster species are cultivated on the French coasts: the Pacific cupped oyster (Crassostrea gigas), which has been introduced from Japan in the 1970's after the disappearance of the Portuguese oyster Crassostrea angulala, and the European flat oyster (Ostrea edulis). During the second half of the 1960's, the latter suffered successively from two diseases caused by protozoan parasites: marteiliosis (also called "maladie des Abers") and bonamiosis. In France, its production dropped from 20,000 tian in the 1960's to 2,000 today. The impact of these two pathogens as well as the exploitation of the remaining stocks show the importance of the conservation of this species, the only natural European species, whose population is more and more fragmented. |

Feeding strategies and digestive capacities can have important implications for variation in energetic pathways associated with ecological and economically important traits, such as growth or reproduction in bivalve species. Here, we investigated the role of amylase in the digestive processes of Crassostrea gigas, using in vivo RNA interference. This approach also allowed us to investigate the relationship between energy intake by feeding and gametogenesis in oysters. Double-stranded (ds)RNA designed to target the two α-amylase genes A and B was injected in vivo into the visceral mass of oysters at two doses. These treatments caused significant reductions in mean mRNA levels of the amylase genes: −50.7% and −59% mRNA A, and −71.9% and −70.6% mRNA B in 15... |

PCR-based DNA markers enable the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European fiat oyster (Ostrea edulis) populations aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Similar patterns of variation were found using allozymes, microsatellites and mitochondrial DNA, aIl which support an isolation by distance model. High variability was observed in the range of diversity between populations using a mitochondrial marker, a reflection of small effective population sizes in sorne locations. A 10fold quantitative difference was observed in Fst,... |

The domestication of Haliotis tuberculata began recently. During this domestication process, abalone may acquire behavioural and physiological traits to become more adapted to their captive environment. These modifications could be the result of intentional selection on production traits, or of unintentional selection due to specific conditions experienced in the farm environment. In order to study this process, progenies of 3 different broodstock origins were studied. Wild abalone, farmed abalone selected for faster growth at 5% selection pressure and randomly selected farmed abalone were used as broodstock. Farmed abalone were the third generation of abalone resulting from systematic mating between wild and farmed broodstock (either males or females were... |

Three successive generations were produced between 2001 and 2003 to assess to what extent genetic variability exists for this trait in juvenile oysters Crassostrea gigas. For each generation, two groups were selected for their high ("R" for resistant) and low ("S" for susceptible) survival. Significant differences of mortality were observed during the first year in Rivière d'Auray (RA) (Brittany-France). However, low and similar mortality were observed for both groups, with no significant difference between groups in the first and second generation in this site, where the first summer is the critical sensitive period with a clear age effect. In Baie des Veys (BDV) (Normandie-France), where mortalities usually mainly affect adults, mortality between... |

This study investigated the effects of age (first or second summer of life) and environment (three study sites on French coasts) on summer mortality in Crassostrea gigas, using three generations of oysters produced during the Morest project. For each generation, two groups of oysters, selected either as 'resistant' (R) or 'susceptible' (S) to summer mortality at the age of 6 months in Rivière d'Auray (RA, South Brittany), were evaluated. In Ronce (Marennes-Oléron Bay) and RA, R oysters had low mortality during their first and second summers, while mortality was higher during the second summer in Baie des Veys (BDV, Normandy). In RA, S oysters exhibited higher mortality during their first summer than during their second, while the opposite result was found... |

Dinoflagellates of the genus Alexandrium are a major cause of harmful algal blooms (HABs) that have increasingly disrupted coastal ecosystems for the last several decades. Microalgae from the genus Alexandrium are known to produce paralytic shellfish toxins (PST) but also bioactive extracellular compounds (BEC) that can display cytotoxic, allelopathic, ichtyotoxic or haemolytic effects upon marine organisms. The objective of this experimental study was to assess the effects of PST and BEC produced by A. minutum upon juvenile great scallops Pecten maximus. Scallops were exposed for one week to two different strains of A. minutum, the first producing both PST and BEC and the second producing only BEC. Escape response to starfish, daily shell growth,... |

Hatchery techniques are now well handled in an increasing number of aquaculture species. However, unintentional effects of domestication often remain poorly documented. In this context, we studied how current hatchery practices may genetically influence larval traits for which significant genetic variation had previously been reported in the Pacific oyster. Two main factors were studied: culling (i.e. discarding the smallest larvae) and rearing temperature (26°C versus 20°C). In parallel, we estimated genetic diversity of broodstock populations sampled in French hatcheries. Finally, we compared larval development and settlement success in the progeny of one of these hatchery broodstock, which had been selected for seven generations, with those of wild... |

Hatchery production of great scallop, Pecten maximus, remains unpredictable, notably due to poor larval survival. Large-scale flow-through systems up to 3500 L have been developed to avoid the use of antibiotics in static systems. Alternatively, small-scale flow-through systems have been successfully applied for oysters but they proved to be unsuitable to rear scallop larvae. By focusing on physical factors presumed to limit P. maximus larval development, this study aimed to optimize great scallop larvae rearing parameters under controlled conditions. First, the influence of aeration on larval performances, energetic metabolism and antioxidant defences were studied both in static and flow-through systems. Aeration depressed larval food intake, regardless... |

Aneuploidy has previously been observed in the Pacific oyster, Crassostrea gigas, and shown to be negatively correlated with growth. Moreover, a significant impact of atrazine exposure has been described in C. gigas, and persistence of that effect has been observed between generations. Evidence of differential chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique. Pairs 1, 5, 9, and 10 are characterized by the loss of 1 chromosome. As restriction enzyme (RE) digestion chromosome banding allows a better identification of chromosome pairs, we used this technique to identify which chromosomes are affected when aneuploidy is increased by exposure to atrazine. The progeny of oysters contaminated by atrazine were... |

The flat oyster Ostrea edulis is the species endemic from European coasts, Atlantic and Mediterranean. It has been exploited since Roman times in Europe. But its production decreased from around 20000 tons in the 1950ies to 1500 tons nowadays because of two successive diseases due to the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer initiated a program of selection to produce families of oysters tolerant to Bonamia. In this context, a further step is to identify QTLs of resistance to this parasite. Therefore, a genetic map is first now being built. The establishment of a genetic linkage map will represent the basement for the mapping of QTLs, with the ultimate objective to implement marker-assisted selection in O.... |

A comparative study of the growth performances in cupped oysters Crassostrea gigas, Crassostrea angulata and their hybrids was conducted from March to October 1999 in a controlled farm with phytoplankton (Skeletonema costatum) input. The individuals, of the same age, were from genitors which had been identified through a marker analysis of the mitochondrial genome (Boudry et al, 1998). As a consequence, two pure offspring were obtained through the crossing of Portuguese oyster C. angulata (AA) genitors and the crossing of Japanese oyster C. gigas (GG) genitors. Two other offsprings were produced through the hybrid crossing of C. gigas female genitors with C. angulata (GA) male genitors on one hand, and C. angulata female genitors with C. gigas (AG) male... |

A comparative study of the growth of the oysters Crassostrea gigas, Crassostrea angulata and their hybrids was carried out in 1999 in system controlled with contribution of the micro alga Skeletonema costatum. The results showed significant differences of the pondéral growth of the four studied progenies. The animals C. gigas had growth rates 1.4 times greater than C. angulata oysters. The hybrid progenies were shown more powerful than Portuguese oysters but are not different from the Japanese oysters. The ecophysiologic study showed that the averages of filtration and oxygen uptake, obtained by means of a three hours recording, cannot differentiate the four studied progenies. On the other hand, the calculation of the FTA (Feeding Time Activity) allowed to... |
